ABSTRACT

Abstract Fructooligosaccharide is used widely in many foods and pharmaceutical industries and produced by using different ways such as extracting it from plants or producing it by using plants and microorganisms enzymes. In a previous study, we extracted Fructosyltransferase (Ftase) enzyme from pineapple residue and produced FOS. In this study, we measured the antagonistic activity of two synbiotics, the first synbiotic containing Lactobacillus acidophilus and the produced FOS, the second synbiotic containing Lactobacillus acidophilus and standard FOS, against pathogenic bacteria (P. aeruginosa, E. coli, S. aureus and B cereus). The results showed that the antagonistic activity of both synbiotic types was very close, as there were no significant differences between them except in the antagonistic activity against S. aureus, there was a significant difference between the synbiotic containing the standard FOS, which was the highest in its antagonistic activity compared to the synbiotic containing the produced FOS in this study. The activity of the fructooligosaccharide (FOS) extracted from pineapple residue was evident in enhancing the activity of the probiotic bacteria (L. acidophilus), which had a major role in the production of acids and compounds that inhibited the pathogenic bacteria. The diameters of inhibition areas in the current study ranged between 19.33-28 mm, and E. coli was more susceptible to inhibition, followed by S. aureus, P. aeruginosa, and B. cereus, respectively.


Resumo O fruto-oligossacarídeo (FOS) é amplamente utilizado em muitos alimentos e indústrias farmacêuticas, e é produzido por meio de diferentes maneiras, como extraí-lo de plantas ou produzi-lo usando enzimas de plantas e microrganismos. Em um estudo anterior, extraímos a enzima frutosiltransferase (Ftase) do resíduo de abacaxi e produzimos FOS. Neste estudo, medimos a atividade antagônica de dois simbióticos: o primeiro simbiótico contendo Lactobacillus acidophilus e o FOS produzido, e o segundo simbiótico contendo Lactobacillus acidophilus e o FOS padrão, contra bactérias patogênicas (P. aeruginosa, E. coli, S. aureus e B. cereus). Os resultados mostraram que a atividade antagônica de ambos os tipos simbióticos foi muito próxima, pois não houve diferenças significativas entre eles, exceto na atividade antagônica contra S. aureus, em que houve uma diferença significativa entre o simbiótico contendo o FOS padrão, que foi o mais alto em sua atividade antagônica, em comparação com o simbiótico contendo o FOS produzido neste estudo. A atividade do fruto-oligossacarídeo (FOS) extraído do resíduo de abacaxi ficou evidente no aumento da atividade da bactéria probiótica (L. acidophilus), que teve papel importante na produção de ácidos e compostos inibidores das bactérias patogênicas. Os diâmetros das áreas de inibição no estudo atual variaram entre 19,33 e 28 mm, e E. coli foi mais suscetível à inibição, seguida por S. aureus, P. aeruginosa e B. cereus, respectivamente.

@#<p style="text-align: justify;"><strong>Background.</strong> Anxiety and depression are becoming increasingly prevalent today and are often aggravated by day-to-day stresses. Because current management strategies are usually accompanied by unpleasant side effects, there is a need to look into alternative treatment regimens - such as prebiotics - that may provide equally effective anxiolytic and antidepressant effects.</p><p style="text-align: justify;"><strong>Objective.</strong> Therefore, the study aims to determine the effect of a combined fructooligosaccharide (FOS) and galactooligosaccharide (GOS) supplemented diet on anxiety and depression levels in mice subjected to Unpredictable Chronic Mild Stress (UCMS).</p><p style="text-align: justify;"><strong>Methods.</strong> Forty male BALB/C mice were subjected to UCMS under a pretest-posttest control group design where the treatment group received prebiotic supplementation throughout the study. Repeated measures ANOVA was run to evaluate between, within, and time interactions of the measured anxiety parameters using the light-dark box test, and depression parameter using the fur coat state assessment.</p><p style="text-align: justify;"><strong>Results.</strong> Results show that (1) the FOS + GOS treatment did not give the treatment group an advantage over the control group during UCMS, (2) both groups grew more anxious and depressed over time, and (3) the treatment group grew more anxious with time in relation to control in terms of the total time spent in the light side.</p><p style="text-align: justify;"><strong>Conclusion.</strong> These imply that the UCMS protocol was successful in inducing stress in mice, but the FOS + GOS regimen failed to provide anxiolytic and antidepressant effects on male BALB/C mice exposed to UCMS.</p>

Aims@#Short-chain fructo-oligosaccharides (scFOSs) are good prebiotics that enhance the growth of probiotic bacteria. The aim of this study is to determine the effect of scFOSs produced by levan hydrolysis using recombinant endo-levanase from B. lehensis G1 on the growth of probiotics isolated from commercially cultured milk drinks. @*Methodology and results@#Two probiotic bacteria, Lactobacillus casei and L. rhamnosus, were isolated from commercially cultured milk drinks. ScFOSs were produced by levan hydrolysis using recombinant endo-levanase from B. lehensis G1. The scFOS and levan (control) were added independently to the growth medium, and the growth rates of the probiotic bacteria were determined. Results showed that the growth rate of L. casei decreased in the presence of levan compared with the control medium but increased by approximately 20% when supplemented with scFOS produced by Levblg1-N28S. Similarly, the growth rate of L. rhamnosus increased by approximately 20% when supplemented with scFOS produced by Levblg1 and Levblg1-N28S. @*Conclusion, significance and impact of study@#The scFOSs produced by the enzymatic hydrolysis of levan using a recombinant endo-levanase from B. lehensis G1 have significant potential as prebiotics because they were able to promote the growth of the probiotic bacteria.

ABSTRACT

Aim To examine weather fructo-oligosaccharides(FOS) and silybin(Sil) could exhibit synergetic effect on treating obesity-associated non-alcoholic fatty liver disease(NAFLD).Methods Seventy mice were randomly divided into two groups:control group (NCD,fed with normal chow diet),the other sixty mice were fed with high-fat diet (HFD) to establish NAFLD model.Seventy days after the establishment of experimental model,the latter group was then randomly subdivided into six groups:model (HFD),Sil (30mg· kg-1),FOS (2 000 mg · kg-1),Sil (30 mg ·kg-1) combined with FOS of high,medium and low dose respectively.The NCD and HFD group were given 0.5% CMC,and the other groups were fed with high-fat diet and given 10 mL · kg-1 by gavage daily,then body weight and food intake were recorded.Fasting blood glucose,insulin,homeostasis model of assessment for insulin resistence index (HOMA-IR) and oral glucose tolerance tests(OGTT) were measured after 120 days.All mice were sacrificed after 130 days,and blood and liver were collected.Levels of TC,TG,ALT,AST in serum were detected,and liver index and pathology were also examined.Results FOS (2 000 mg · kg-1) showed obvious synergism for Sil-mediated attenuation of levels of TC,TG,ALT,AST in serum,fasting blood glucose,insulin,HOMA-IR,OGTI curve,liver index and pathology,but FOS (4 000 mg · kg-1) could not bring superiority with a double dose,except for its improvement in body weight of mice with NAFLD.Conclusions In the treatment of NAFLD,FOS exhibits synergetic effect with Sil.This agent might be a potent candidate for obesity and NAFLD prevention,through modulating the composition of gut microbiota.

ABSTRACT

Um experimento foi realizado para avaliar o efeito da utilização de diferentes promotores de crescimento sobre o metabolismo de frangos de corte em crescimento. Foram avaliadas duas dietas, uma atendendo às exigências nutricionais de frangos de corte (normal) e uma dieta reduzida, com 5% a menos da exigência nutricional (reduzida), suplementadas ou não com antibióticos e prebióticos. Foram utilizados 180 pintos de corte criados em galpão convencional, com cama de maravalha, até os 21 dias de idade. Em seguida, foram divididos em 36 gaiolas, distribuídos em delineamento experimental inteiramente ao acaso, utilizando método tradicional de coleta de excretas. Houve interação (P<0,05) entre dieta e promotores de crescimento para os coeficientes de digestibilidade ileal da matéria seca (CDIMS), da proteína bruta (CDIPB) e do extrato etéreo (CDIEE). A dieta reduzida contendo antibiótico apresentou menor CDIMS. A dieta normal contendo antibiótico apresentou maior CDIPB (P<0,05), enquanto a dieta reduzida contendo prebiótico apresentou melhor CDIPB e CDIEE (P<0,05). Os valores dos coeficientes de digestibilidade total da matéria seca (CDTMS), da proteína bruta (CDTPB) e do extrato etéreo (CDTEE) apresentaram interação (P<0,05) entre dietas e promotores de crescimento, em que a dieta reduzida contendo antibiótico apresentou menor CDTMS e CDTEE. A dieta normal apresentou maiores valores de energia metabolizável aparente (EMA) e EMA corrigida para balanço de nitrogênio (EMAn) (P<0,05). Concluiu-se que, ao se reduzir a exigência nutricional da ração, a digestibilidade dos nutrientes foi reduzida, no entanto os óleos essenciais promoveram aumento nos coeficientes de digestibilidade ileal da matéria seca, da proteína bruta e do extrato etéreo.


An experiment was conducted to evaluate the effect of using different growth promoters on the metabolism of growing broilers. We evaluated two diets, one meeting the nutritional requirements of broilers (normal) and another reduced diet with 5% less than the nutritional requirement (reduced), supplemented or not with antibiotics and prebiotics. A total of 180 broiler chicks reared in shed masonry and wood shaving bedding until 21 days of age. The animals were divided into 36 cages and distributed in a completely randomized design, using a traditional method of excreta collection. There was an interaction (P<0.05) between diet and growth promoters for ileal digestibility coefficient of dry matter (IDCDM), crude protein (IDCCP) and ether extract (IDCEE). The reduced diet containing antibiotics had lower IDCDM. The normal diet containing antibiotic had higher IDCCP (P<0.05), whereas the reduced diet containing prebiotic showed better IDCCP and IDCEE (P<0.05). The values of total digestibility coefficient of dry matter (TDCDM), crude protein (TDCCP) and ether extract (TDCEE) showed interaction (P<0.05) between diets and growth promoters, where the reduced diet containing antibiotics had lower TDCDM and TDCEE. A normal diet had higher apparent metabolizable energy (AME) and AME corrected for nitrogen balance (AMEn) (P<0.05). It was concluded that when the nutritional requirement of diet was reduced, the nutrients digestibility was also reduced, however essential oils caused an increase in ileal digestibility of dry matter, crude protein and ether extract.

ABSTRACT

OBJECTIVE To investigate chemical properties of a fructooligosaccharide (ABP-50-FOS)separated from Achyranthes bidentata and immune response in mice immunized H1N1 influenza vaccine. METHODS The methods of GPC,CE,IR and NMR were used to study chemical properties of ABP-50-FOS. BALB/c mice were immunized intramuscularly twice with H1N1 influenza vaccine (3 μg)plus ABP-50-FOS(200 μg)each mouse. The serum total antibody titer and its isotypes titers were analyzed by ELISA. The populations of CD4+,CD8+,CD3+and CD19+lymphocytes were deter?mined by flow cytometry. The proliferation activities of spleen T and B lymphocytes were determined with MTT method. The levels of cytokines interferon-γ(IFN-γ),tumor necrosis factor-α(TNF-α),inter?leukin-4(IL-4),IL-12 and NO were measured by ELISA kits. RESULTS ABP-50-FOS was a fructooli?gosaccharide with moleculer mass 1885 u. Its bone linkages contained 1,2-and 1,6-fructose residues. ABP-50-FOS could induce high specific-IgG,IgG1,IgG2a,IgG2b and IgM titers after immunization with H1N1 influenza antigen twice(P<0.01). ABP-50-FOS significantly elevated the percentage of CD3+,CD4+ and CD8+ spleen lymphocytes and IFN-γ secretions(P<0.01)in vitro. It also stimulated peritoneal macrophage of mice and DC2.4 dendritic cells to produce TNF-αand IL-12p70 respectively (P<0.01). However,ABP-50-FOS inhibited secretions of NO in macrophage. CONCLUSION The fruc?tooligosaccharide ABP-50-FOS separated from A. bidentata can exhibit strong adjuvant activity for H1N1 influenza vaccine.

ABSTRACT

This present study focuses on the microbial production of Fructosyl Transferase (FTase) and the production of Fructooligosaccharides(FOS) by transfructosylation using this enzyme. Fructooligosaccharides are industrially produced from sucrose by microbial enzymes with transfructosylating activity. Fructosyltransferase is produced intracellulraly by fungi. In the present study fungus Aureobasidium pullulans was used to produce FOS and effect of different physical and chemical parameters on FOS production were studied. The analysis of the product was performed by High Pressure Liquid Chromatography (HPLC).

ABSTRACT

Fructooligosaccharide (FOS) is a prebiotic, becoming apparent for its therapeutic role for diseased conditions like CHD, hypertension, diabetes and obesity. CHD and hypertension are one of the most prevalent NCD’s encountered in diabetic adults. Present study aims to assess the effect of FOS supplementation in type 2 diabetic adults on their lipemic, biophysical parameters and gut microflora parameters. A cross-sectional study was designed with 65 adult type 2 diabetics enrolled from Health clinic of The M.S. University of Baroda, Gujarat. All the subjects were randomly divided into two groups control and experimental. The experimental group was given 10 g of fructooligosaccharide and compared with the controls for lipemic parameters, hypertension and fecal counts in terms of lactic acid bacteria, bifidobacteria and enteric pathogen. Eight week supplementation of FOS resulted in an appreciable reduction in serum TC, TG and LDL levels by 10%, 4.9% and 7.8% respectively. A significant reduction (p<0.05) in systolic blood pressure was also observed as a result of FOS supplementation in the experimental group. A decline was seen in TC/HDL, LDL/HDL and non-HDL by 10.4%, 7.6% and 6.6% respectively (p<0.05, p<0.001). Gut microbiota values exhibited a significant increment in fecal log10 counts of Lactic acid bacteria and bifidobacteria by 9.3% and 10.9% respectively (p<0.001) while a significant reduction by 4.8% (p<0.001) was observed for Enteric pathogen. These outcomes revealed an efficacy of FOS in reducing serum lipid parameters, systolic BP and improving the gut microbiota in type 2 diabetic adults.

ABSTRACT

Prebiotics are the non-digestible fibers which have the potential to exert a positive effect on human health. They serve as a substrate for microflora in the large intestine and are known to improve overall gut health. Fried foods contribute greatly to the snack foods industries and are specially liked for their crispy texture and unique flavor. Addition of fructooligosaccharides in fried food products has a potential to be developed into a therapeutic food. Hence, the effect of incorporating fructooligosaccharide on sensory qualities of fried products namely vegetable paratha (Shallow fried) and lilva kachori (Deep fried) was investigated. Fructooligosaccharides was added to the one serving of vegetable paratha and lilva kachori’s stuffing at 10 g, 15 g and 5 g, 10 g, and 15 g respectively and were compared with the standard recipes. Three successive trials were conducted for screening the panelist through threshold test. Sensory evaluation was carried out at nine point hedonic scale test for the two products in triplicates. An internal panel of 25 semi trained members evaluated the products for color and appearance, mouthfeel, texture, taste, after taste, and overall acceptability. The results revealed a non-significant reduction in most of the sensory attributes for vegetable paratha, whereas, a significant reduction was observed in texture scores (p<.05). A significant reduction (p<.001) was found in all sensory attribute scores of lilva kachori. Addition upto 5 g fructooligosaccharide did not altered sensory qualities of lilva kachori. The most affected attribute for vegetable parantha was texture (8.6 percent reduction) and for lilva kachori it was color and appearance. Thus it can be concluded that fructooligosaccharides can be incorporated in vegetable paratha upto 15 g and upto 5 g in lilva kachori. An addition of fructooligosaccharide in stuffed fried snacks may serve to fulfill consumer demand for health foods.

ABSTRACT

This study evaluated the effects of prebiotics on fermentation profile and growth of Lactobacillus acidophilus, Lactobacillus bulgaricus, Lactobacillus rhamnosus, and Bifidobacterium lactis in co-cultures with Streptococcus thermophilus. Acidification rate and viability were positively influenced by the co-culture with B. lactis and by both inulin or oligofructose in low fat milk.

ABSTRACT

The objective of the present study was to study the effect of the variables of the osmotic dehydration process on sliced Fuji apples (Malus domestica) using a 2 x 3² factorial design. The variables studied in the apple slices were the pretreatment (blanching or acidification), the temperatures (30, 45 and 60ºC) and the FOS concentration (40%, 50% and 60% m/v) of the osmotic solution. There was no difference among the pretreatments for the water activity and titratable acidity. The slices pre-treated by the acidification presented less enzymatic browning (greatest luminosity L* value) combined with a greater soluble solid contents (thus, this treatment was selected). Treatments T4 (45ºC and 40% m/v) and T7 (60ºC and 40%m/v), using the acidification presented responses within the recommended standards and FOS were validated by the repetition.

ABSTRACT

O excesso de Fe no organismo gera espécies reativas de oxigênio (ERO) que são potencialmente tóxicas. Entretanto, a magnitude dos efeitos da exposição à moderada sobrecarga de Fe e da sua interação com facilitadores e/ou inibidores da absorção mineral não é conhecida. O objetivo deste trabalho foi avaliar esses efeitos e a sua interação com fruta nos e/ou fitato (facilitadores e inibidores da absorção de Fe, respectivamente) nos índices séricos do estado nutricional em Fe, no perfil dos lipídeos séricos e em parâmetros do metabolismo hepático e ósseo. Para o experimento foram utilizados 34 ratos machos Wistar, pesando inicialmente 49,3 ± 3,9g, alojados individualmente em gaiolas de aço semimetabólicas por 92 dias. Uma dieta AIN-93G (Dieta 1: Grupo Controle) e quatro dietas AIN93G modificadas foram usadas para o estudo, com as seguintes características: Dieta 2: sobrecarga moderada de ferro com 550mgFe/kg de ração (Grupo SBC); Dieta 3: sobrecarga moderada de ferro + 18% de farinha de yacon (Grupo SBC+FY); Dieta 4: sobrecarga moderada de ferro + 0,6% de fitato (Grupo SBC+FIT); Dieta 5: sobrecarga moderada de ferro + 18% de farinha de yacon + 0,6 % de fitato (Grupo SBC+FY+FIT). Os resultados demonstraram que a moderada sobrecarga de Fe ou a sua interação com farinha de yacon e/ou fitato não alterou os índices séricos do estado nutricional em Fe. Ocorreu aumento na atividade sérica da AST apenas no grupo SBC (p=0,055). Nos grupos SBC e SBC+FY houve diminuição na concentração do colesterol sérico (p=0,002) e, apenas no grupo SBC+FY+FIT, diminuição da concentração sérica do VLDL. No fígado, houve aumento significativo (p=0,002) da concentração de Fe não-heme nos grupos IO (+83%) e SBC+FIT (+117%) e, em todos os grupos SBC, na atividade da enzima GPx (p=0,000). A atividade da CAT foi menor (p=0,036) apenas para o grupo SBC+FY+FIT. Em todos os grupos SBC ocorreu significativo aumento nos depósitos de hemossiderina em torno das células de Kupffer (p=0,000). Houve aumento na apoptose em todos os grupos SBC, com os grupos SBC+FY e SBC+FY +FIT apresentando o maior número de corpúsculos apoptóticos/área (+405% e +342%, respectivamente) (p=0,000). Não houve alteração nos parâmetros relacionados ao metabolismo ósseo. No grupo SBC+FY houve significativo aumento na absorção aparente de Ca (p<0,05). Conclusões: A moderada sobrecarga de Fe não alterou os índices séricos do estado nutricional em Fe, mas resultou em alterações no tecido hepático e no perfil dos lipídeos séricos. Com exceção do perfil de lipídeos séricos, no qual apenas o fitato pareceu exercer efeito protetor, nos demais parâmetros avaliados a interação com farinha de yacon rica em fruta nos e/ou fitato reverteu parcial ou totalmente as alterações induzidas pela moderada sobrecarga de Fe


Excess Fe in the organism generates potentially toxic reactive oxygen species (ROS). However, the magnitude of the effects of a moderate Fe overload and its interaction with factors which inhibit or facilitate mineral absorption is not known. The aim of the present work was to evaluate such effects and their interaction with fructans and/or phytate (compounds which facilitate and inhibit Fe absorption, respectively) on serum iron status indices, on the profile of serum lipids and on hepatic and bone metabolism parameters. In the experiment, thirty-four male Wistar rats initially weighing 49,3 ± 3,9g were used. The rats were housed in individual stainless-steel wire-mesh cages for 92 days. An AIN-93G diet (Diet 1: Control Group) and four modified AIN-93G diets were used in the study. The modified diets presented the following formulations: Diet 2: moderate Fe overload with 550mgFe/kg diet (IO Group); Diet 3: moderate Fe overload + 18% yacon flour (IO-YF Group); Diet 4: moderate Fe verload + 0.6% phytate (IO-Phy Group); Diet 5: moderate Fe overload + 18% yacon flour + 0.6% phytate (IO-YF-Phy Group). The results demonstrated that a moderate Fe overload or its interaction with yacon flour and/or phytate did not alter the serum iron status indices. An increase in the serum AST activity was observed only in the IO group (p=0,055). In the IO and IO-YF groups, there was a reduction in the serum cholesterol concentration (p=0,002) and a reduction in the serum VLDL concentration was observed only in the IO-YF-Phy group. In the liver, there was a significant increase (p=0,002) in non-heme Fe concentration in the IO (+83%) and IO-Phy (+117%) groups. Also, GPx activity was significantly increased (p=0,000) in all IO groups. CAT activity was lower (p=0,036) only in the IO-YF-Phy group. A significant increase in hemosiderin deposition around Kupffer cells was observed in all IO groups (p=0,000). Apoptosis was increased in all IO groups, whereas the IO-YF and IO-YF-Phy groups showed the largest number of apoptotic bodies/area (+405% and +342%, respectively) (p=0,000). There was no alteration in the parameters related to bone metabolism. In the IO-YF group, there was a significant increase in Ca apparent absorption (p<0,05).Conclusions: The moderate Fe overload did not alter the serum iron status índices, but led to alterations in the hepatic tissue and in the profile of serum lipids. Except for the profile of serum lipids where only phytate seemed to have a protective effect, in the other evaluated parameters the interaction with yacon flour rich in fructans and/or phytate partially or totally reversed the alterations induced by the moderate Fe overload

ABSTRACT

This study was carried out to investigate the efficacy of sn-2 palmitic acid-fortified vegetable oil (Sn2PA) on calcium absorption and to confirm the synergistic effects of fructooligosaccharide on calcium absorption. Male SD rats were fed 6 kinds of casein based diets containing vegetable oil (control), sn-2 palmitic acid-fortified vegetable oil (Sn2PA) and Sn2PA with fructooligosaccharide(Sn2PAFO) in two levels of calcium (normal 0.5% and high 1.0%) for 3 weeks. Total lipids, cholesterol, triglyceride and calcium in blood were measured. Feces were collected using cages for 4 days. Serum concentrations of total lipids and calcium were not significantly different among groups. However, serum triglyceride was significantly decreased by fructooligosaccharide supplementation regardless of dietary calcium level. The lipid absorption was not significantly different among experimental groups. Calcium absorption was significantly higher in Sn2PAFO group than other groups. Calcium solubility of intestine was increased by sn-2 palmitic acid supplementation. These results suggest that sn-2 palmitic acid and fructooligosaccharide supplementation could be beneficial for baby foods including infant formula, with regard to increasing absorption of calcium by more soluble calcium in the small intestinal content.

ABSTRACT

We investigated the combinatorial effects of different doses of dietary soy isoflavones (SI) and fructooligosaccharide (FOS) in a rat model of colon cancer. We hypothesized that increased bioavailability of SI metabolites due to dietary FOS may increase production of bioactive equol and affect colon carcinogenesis in a dose-dependent manner. Sprague-Dawley male rats were injected with 12-dimethylhydrazine (DMH) and were provided experimental diets that contained 0, 10, 50, 150, or 500 mg SI per kg of diet and 6% FOS for 12 weeks. The number of aberrant crypt foci (ACF) and the expression of cyclooxygenase-2 (COX-2) in colonic tissues were significantly decreased in the 6% FOS-fed groups compared to the control group. Gut transit time and fecal pH were significantly lower, and fecal concentrations of bifidobacteria were increased with 6% FOS. However, dietary SI supplementation in combination with 6% dietary FOS did not affect ACF formation or COX-2 expression. Plasma equol concentrations were dose-dependently increased by supplementation of SI up to 500 mg/kg of diet. In conclusion, SI supplementation up to 500 mg/kg of diet appeared to have no additive beneficial effects in rats with chemically-induced colon cancer that were fed 6% FOS, although plasma equol was dose-dependently increased.

ABSTRACT

OBJETIVO: Verificar o efeito bifidogênico do frutooligossacarídeo nos pacientes com neoplasias hematológicas submetidos a quimioterapia. MÉTODOS: Trata-se de um estudo clínico randomizado duplo cego, desenvolvido na Unidade de Transplante de Medula Óssea do Centro de Pesquisas Oncológicas de Florianópolis, o qual envolve 25 pacientes divididos em 2 grupos que receberam, por 15 dias, 12g de frutooligossacarídeo (n=14) ou placebo (maltodextrina) (n=11). Foram avaliados a quantidade de bifidobactérias e os valores de pH fecal antes e após a suplementação. RESULTADOS: Observou-se na população estudada o predomínio do sexo masculino (72 por cento) e a idade média de 34 anos. O grupo suplementado apresentou um aumento significante na quantidade de bifidobactérias (p<0,05) e o pH fecal não foi alterado em nenhum dos grupos. CONCLUSÃO: Verificou-se que a suplementação aumentou a quantidade de bifidobactérias, interferindo na composição da microbiota intestinal, e que não houve alteração do pH fecal.


OBJECTIVE: To verify the bifidogenic effect of fructooligosaccharides in patients with hematological neoplasia submitted to chemotherapy. METHODS: This is a clinical, randomized, double-blind study done in the Bone Marrow Transplant Unit of the Oncology Research Center of Florianopolis. It involved 25 patients divided into 2 groups who received 12g of fructooligosaccharides (n=14) for 15 days or placebo (maltodextrin) (n=11). The amount of bifidobacteria and the values of fecal pH before and after supplementation were investigated. RESULTS: Most of the studied population was male (72 percent) and the mean age was 34 years. The group that received supplementation presented a significant increase in the amount of bifidobacteria (p<0.05) and fecal pH remained unchanged in both groups. CONCLUSION: Supplementation increased the amount of bifidobacteria, interfering in the composition of the intestinal flora, but fecal pH was not affected.

ABSTRACT

Low glycemic index (GI) foods have been reported to be very important in the control of blood glucose levels in diseases such as diabetes mellitus and obesity, which are becoming more prevalent in Korea. Bread consumption at breakfast and as a snack is also on the rise. To provide low GI alternatives, breads supplemented with dietary fibersfructooligosaccharide (FOS), ground whole rye, and 2 types of resistance starch (RS2 and RS4)-were developed. The GIs for these breads were evaluated with 13 healthy college students (M6, F7) and sensory tests were done. Four kinds of breads were made through the modification of a basic recipe for white bread (the control, B) as follows: for Bf, the entire amount of sugar was replaced with FOS. For Ryef, 50% of the white wheat flower was replaced with roughly ground wholerye (20 mesh: 12-20 mesh = 1 : 3) in addition to replacing sugar with FOS. For RS2f and RS4f, 20% of the white wheat flower was replaced with RS2 or RS4, in addition to replacing sugar with FOS. The overall preference score of Bf was similar to that of B, while those of Ryef, RS2f and RS4f were lower than that of B, but showed the acceptable degree of the overall preference. The glycemic indices of Ryef and RS2f were 46.1 and 45.9 respectively, which were significantly lower than the GI of B, 67.8. The GIs of Bf and RS4f were, however, 66.7 and 80.5 respectively, showing no significant difference compared to B. The glycemic loads for a 30 g serving were 9.5, 5.9, 6.2, 11.0 and 9.0 for B, Bf, Ryef, RS2f and RS4f, respectively. In conclusion, addition of RS2 or roughly ground whole rye to the dough formula significantly lowered the GI. Since the preferences shown for those two breads were acceptable, they may be recommended as a substitute for white bread for persons who need blood glucose management. More studies on the bread making process are required to improve preference and acceptance. Although GI lowering effects for F and RS4 were not found in this study, further studies are needed to verify their effects.

ABSTRACT

Fructooligosaccharide (FOS) is a typical prebiotic agent. A randomized, double-masked, placebo-controlled study was performed to evaluate the prebiotic effect of daily intake of an isotonic solution containing FOS on body weight gain and the reduction of diarrhea in children in an urban slum in Bangladesh over six consecutive months. We enrolled a total of 150 children, aged 25-59 months. Sixty-four children in the FOS group received 50 mL of isotonic solution with 2 g of FOS added, and 69 children in the placebo group were given an identical solution with 1 g of glucose added, once a day. The measurement of body weight was carried out every other day; height and arm circumference were measured once a month; and the children‘s mothers were interviewed to obtain data about diarrhea, the consistency and constitution of stool, other symptoms, and antibiotic treatment. As a result, the body weight gain during the six-month period was 0.86±0.55 kg in the FOS group and 0.89±0.48 kg in the placebo group, while the increase in height and arm circumference were not significantly different between the two groups. The number of diarrhea episodes during the six-month period was not significantly different. A significant reduction in the duration of diarrhea days and of duration per episode was observed in the FOS group (<i>p</i>=0.039 and <i>p</i>=0.008, respectively). In conclusion, daily intake of FOS was associated neither with the children‘s growth nor with the number of diarrhea episodes, but a significant reduction in the duration of diarrhea days was observed. Further studies are needed to confirm the effects of FOS by changing the doses and eliminating the influence of antibiotics.

ABSTRACT

The purpose of this study was to elucidate effects of fructooligosaccharide on gastrointestinal tract and blood lipids of rats when this was supplied as purchased condition or oligosaccharide containing sponge cake. Male Sprague-Dawley rats were assigned to one of 3 treatments 1) control diet 2) 7.5% fructooligosaccharide containing diet (FOS diet) 3) lyophilized sponge cake powder containing diet (FOS-C diet). The sponge cake was made with fractooligosaccharide which replaced 40% of its surose, and the final concentration of fructooligosaccharide in FOS-C diet was 7.5%. Cecal and fecal water contents, amount of cecal content, and cecal wall weight were higher from fructooligosaccharide consumption, whereas total gut transit time was longer in rats consuming fructooligosaccharide compared with those fed control diet. Cecal and fecal pH were lower in FOS and FOS-C groups than in control group. Total cecal SCFA pools were higher from ingesting fructooligosaccharide containing diets compared with control diet. Serum triglyceride levels were lower in rats fed FOS and FOS-C diet than those fed control diet, while serum cholesterol levels were unaffected by treatment. Therefore the effects of fructooligosaccharide in sponge cake on serum lipids and gastrointestinal tract were similar to those of intact fructooligosaccharide. Also, adding 7.5% of FOS accompanied diarrhea symptom which suggests some precaution are needed when using FOS.

ABSTRACT

Objective: To study the effects of dietary fructooligosaccharide (FOS) on cholesterol metabolism in finishing pigs. Methods: A total of 96 finishing pigs (Duroc?Landrance?Jia) were randomly alotted to four groups each of which was replicated three times with eight pigs per replicate. The groups received the same basal corn-soybean meal diet and FOS was added to the basal diet at 0,0.5%,1 0%,1.5% at the expense of corn, respectively. At the end of the feeding trial, eight pigs with similar body weights from each treatment were slaughtered. Results: (1) Supplementation with FOS significantly improved average daily gain and feed efficiency. (2) Pigs fed 1.0% and 1.5% FOS had significantly lower serum TC and LDL-C concentrations and greater HDL-C/LDL-C ratio compared with the control. Pigs fed 1.5% FOS had significantly lower serum VLDL-C and ApoB concentrations and Apo B/Apo A-1 ratio. (3) Supplementation with 1.0% and 1.5% FOS significantly decreased the activities of HMG-CoA reductase. (4) Supplementation with 1.5% FOS significantly increased the colonic acetate and butyrate and plasma acetate. Supplementation with 0.5%,1.0% and 1.5% FOS significantly increased colonic propionate. (5) Fecal cholesterol excretion was significantly greater in the pigs fed 1.0% and 1.5% FOS and fecal bile acid excretion was significantly greater in the pigs fed 1.5% FOS than the control. Conclusion: FOS may improve cholesterol metabolism by alteration in the absorption and synthesis of cholesterol resulted from the changes in colonic fermentation, and by increase in the fecal excretion of cholesterol and bile acid.
